چکیده

S-adenosylmethionine, known for some time to be the biological methyl donor in mammalian enzymatic transmethylation reactions (1,2), has more recently been implicated in other transalkylation reactions in bacteria and yeast (3-6). The enzyme system catalyzing the condensation of ATP and methionine to form AMe’ has been found in a variety of tissues and organisms, and has been purified from two sources and investigated in detail (7, 8). The broad distribution of these enzymes suggests that AMe likewise has widespread distribution and lends importance to a study of its further metabolism. The present paper reports the presence of an enzyme in baker’s yeast which decomposes AMe to 5’-methylthioadenosine and a-aminobutyrolactone (Equation 1; A = adenine, R = ribose) .

Recent Advances in Radical SAM Enzymology: New Structures and Mechanisms

The radical S-adenosylmethionine (SAM) superfamily of enzymes catalyzes an amazingly diverse variety of reactions ranging from simple hydrogen abstraction to complicated multistep rearrangements and insertions. The reactions they catalyze are important for a broad range of biological functions, including cofactor and natural product biosynthesis, DNA repair, and tRNA modification. Generally con...

The enzymatic decomposition of S-adenosyl-L-methionine.

The role of S-adenosyl-n-methionine as a methyl donor has been investigated in mammalian systems (1) and in microbial metabolism (2-4). In addition, the compound is involved in the formation of spermidine from putrescine (5). Information concerning the stability of S-adenosylmethionine in various enzymatic systems is a necessary prerequisite for studies concerning its role in group transfer. In...

Spermidine Biosynthesis in Saccharomyces cereuisiae

We have cloned and sequenced the Saccharomyces cerevisiae gene for S-adenosylmethionine decarboxylase. This enzyme contains covalently bound pyruvate which is essential for enzymatic activity. We have shown that this enzyme is synthesized as a M, 46,000 proenzyme which is then cleaved post-translationally to form two polypeptide chains: a fi subunit (Mr 10,000) from the amino-terminal portion a...

The preparation of S-adenosylmethionine.

Although the enzymatic synthesis of S-adenosylmethionine was accomplished several years ago (l), there have, as yet, been few reports of experiments with this compound. The main reason for the paucity of data appears to be the difficulty of obtaining S-adenosylmethionine in adequate quantities at a reasonable expenditure of time and starting material. Cantoni (2) has prepared S-adenosylmethioni...

The antiviral protein viperin is a radical SAM enzyme.

Viperin, an interferon-inducible antiviral protein, is shown to bind an iron-sulfur cluster, based on iron analysis as well as UV-Vis and electron paramagnetic resonance spectroscopic data. The reduced protein contains a [4Fe-4S](1+) cluster whose g-values are altered upon addition of S-adenosylmethionine (SAM), consistent with SAM coordination to the cluster. Incubation of reduced viperin with...
